摘要

为探讨缝隙连接蛋白 43(Connexin43,Cx43)在人脑胶质瘤中的表达水平及其与肿瘤恶性程度的相关性。采用 HE 染色、免疫组化法确定胶质瘤性质等级。标本分 5 组:正常脑组织组、WHOI 级胶质瘤组、WHOII 级胶质瘤组、 WHOIII 级胶质瘤组、WHOIV 级胶质瘤组。通过 Ki-67 的染色反应肿瘤细胞增殖情况。采用 Western Blot 技术检测 Cx43 蛋白的表达。Cx43 蛋白在正常脑组织中和在 WHOI 级胶质瘤中表达均较高,但二者差异无统计学意义 (P>0.05),在 WHOI-IV 级胶质瘤组中表达差异性有统计学意义,呈现逐渐减少的趋势。Cx43 蛋白表达随着胶质瘤恶性程度的增 高而降低,与细胞增殖和肿瘤组织病理学分级呈负相关。由此可知,Cx43 蛋白与肿瘤细胞活性和胶质瘤恶性进展密 切相关。

Abstract

To explore the relationship between the expression of gap Connexin43 (Cx43)level in human gliomas and correlation with tumor grade.HE staining and immunohistochemical method was used to determine the glioma grades.Specimens were divided into 5 groups:normal,WHOI grade glioma,WHOII grade glioma,WHOIII grade glioma,WHOIV grade glioma.Ki- 67 staining to examine cell proliferation.Western Blot to detect expression of Cx43.The result showed that the Cx43 was high expressed in normal brain and WHOI in gliomas,but difference was no significance (P>0.05),while there were significantly differences between the other WHOI- IV groups (P<0.001),and it showed a diminishing trend with reduced glioma malignant degree.The results indicate that Cx43 exprssion is negative correlated with tumor cells proliferation and pathological grade of tumor tissue,and closely related to the activity of tumor cells proliferation and the maglinant progression of glioma.
